Behind the Camera, Checking the Ego

David O. Russell does not have the reputation of an evenhanded filmmaker. Depending on whom you talk to, Mr. Russell, whose work includes the Persian Gulf war satire "Three Kings" and the existential corporate comedy "I ♥ Huckabees," is quixotic, combative and occasionally brilliant. for his latest film, "The Fighter," a passion project brought to him by his frequent star Mark Wahlberg, Mr. Russell takes on the true story of Micky Ward and Dicky Eklund, brothers and struggling boxers from Lowell, Mass., near where Mr. Wahlberg grew up. He plays Micky, the quieter but more successful of the two, and Christian Bale , in an outsize, twitchy performance that is generating Oscar talk, plays Dicky, a recovering drug addict and a wiry charmer.

The film, which opens in new York, Los Angeles and Boston on Friday, focuses on several fights, which Mr. Russell filmed in period style, using the same cameras and cameramen that HBO did when it broadcast them. In an interview at the Four Seasons restaurant in Manhattan, Mr. Russell spoke about his technique, his career and why surviving a few rounds in a boxing ring is good preparation for Hollywood. these are excerpts from that conversation.

Q.

The film opens with a documentary-style interview with Micky and Dicky that exposes their relationship, but that wasn't in the original script?

A.

[It was] a discovery in the editing room. We had no time in the schedule. You shoot these interviews as you go, throw them in the frames: Tell me about this. Talk about this.

Q.

It was totally unscripted?

A.

We had worked on the characters. Christian was in character nonstop, and mark, coming from the John Garfield school, is always kind of in character, because it's always him in some way. I pretend to be the HBO interviewer, and I would say, tell me when you started fighting. Then Christian would go down the road, and then I would say, ‘No, say this.' I would just start throwing lines at him that we would make up on the spot, and we got that gold where he cried at the end. That's like a cinematic haiku.

Q.

Tell me about working with Christian. He has a reputation as an intense actor.

A.

Christian is a serious guy, and by that I mean he's a sincere guy, so when he commits to something, you can feel how much sincerity there is. And we both talked about wanting to come from a place of love for Dicky, that's what made it interesting. We had to figure each other out the first couple of weeks, but once we found a rhythm we were very, very comfortable. He does his thing. He goes off to his workshop, and he comes back with his shaved hairline and his bones sticking out.

He listened to the music of Dicky and the movement of Dicky. Dicky is basically a black guy, like a Muhammad Ali type of black guy who never stops moving and talking, which was very liberating to Christian, because he is a very quiet person. And then there was a question of calibrating it. He came out of the gate with his creature, and I would have to say: “You've got to bring it down a little bit, and it's a matter of trusting me, understanding that you're still very much Dicky, you're just a little too big right now.” Ideally in any director-actor relationship that's what you want: is this too much? is this too little?

Q.

Mark trained for four years to play a boxer, and you shot all the fight scenes in just the first three days. how did you choreograph them?

A.

We picked our rounds that we loved of Micky's fights. We would have them up on YouTube while we were shooting and I'd go, 'Wait, look, that isn't exactly right.' We had to make sure we had the angles for the right hits. We had 79 hours of footage for only three days. We still had more footage than we knew what do with, so the real job was to find the most dynamic rendition and cinematic rendition of the drama of these particular fights, each one very different.

Q.

You shot on location in Lowell, in a blue-collar community, and you depict drug addiction and crime. how do you avoid moralizing in that context?

A.

I think the tone is really set by the real people themselves, because the way they take that stuff in stride is how the film needed to take it in stride. Meaning it's been part and parcel of their lives for a long time, going back to their uncles and their fathers, they're all in and out of jail. Micky Ward's dad was in jail with Mark's dad. That's how they are. They're beautiful, big-hearted people, you would think they were venal in some way and they're not. They talk about it in an unflinching way, just the way they take a punch.

That's the most beautiful thing that I like about boxing: You can take a punch. the biggest thing about taking a punch is, your ego reacts and there's no better spiritual lesson than trying to not pay attention to your ego's reaction. That's what takes people out of the fight half the time. They get hit and half the reaction is, your ego is saying, “I cannot believe that person just lit me up, how humiliating.” And what a fighter has to do and what Micky does and what these guys do, whether it's a prison thing or a crime or a drug episode, is they kind of just go [He mimes ducking and getting up].

Q.

Let me ask you to draw a parallel between being able to take a punch and your career as a filmmaker. You're no stranger to difficult shoots and things that have battered your reputation. [An altercation with George Clooney on the set of "Three Kings," for example.] is that part of your interest in learning how to bounce back?

A.

Absolutely. That's where you really have to show what your character is. That's why I love Bill Clinton. I just love people who can take a punch and pick themselves up and come back. That's why I found Micky so inspiring, and Dicky. I had a few humbling years, and those years made me a better filmmaker. I write better now, and I see better now cinematically. There's a lot less deliberation and more instinct and 'Get to it, Jack.' Cinematically, your hunger gets deeper. this shoot was not a hard shoot, as short as it was, because we're all grateful to be there, me, for all the reasons I'm telling you right now. I loved the people, I almost wish there was a TV series that we were still shooting. I like being with those people, because they know how to live. They are the roofers and the pavers, they're very alive and very unpretentious and very open.

Q.

Unless you're an interloper.

A.

That's where mark helped. mark is from almost virtually the same family of nine children [there are nine children in the Ward-Eklund clan], and so that helped a lot, and also I was an organizer up in those parts before I became a filmmaker. I was a community organizer for housing projects in Lewiston, me., which is very much like Lowell, and in the South end of Boston. my first films were documenting housing conditions and the people in them in Lewiston in the early '80s and the same thing then in the South end of Boston for Central American immigrants, writing about people who are getting racially harassed in South Boston, probably by mark and his friends. it was coming up on the time when mark went to prison.

Q.

You've worked with mark before. Tell me about that relationship, because he's not necessarily an actor I expect you to have a kinship with.

A.

We really liked each other and we crack each other up and we can be very authentic with each other. [But] I know what you mean. You almost would expect me to be with a more voluble — like a Cusack, or who’s the version of that now? I’ve written for Vince [Vaughn] a lot, Vince is a big talker. I love listening to Vince talk. a lot of it is musical to me or rhythmic, and Dustin Hoffman taught me that that’s O.K. I used to think that was a bad — that was my dirty little secret.

Like as a way of directing and as a way of feeling a scene, I feel rhythmically or musically, I feel the music, and I guess Quentin [Tarantino] does this as well, he talks about it as music, and people made fun of that. And Dustin said, no, that is the character, that is the person, the musicality of how you’re feeling it in the scene, so do tell us that.

Q.

A lot of your other films are satirical, and here you're really striving for authenticity in everything, from the camera work to your depiction of the characters. is that a change that you will continue?

A.

I think for sure. You can't go wrong. It's really, really simple. the closer you stay to emotional authenticity and people, character authenticity, the less you can go wrong. That's how I feel now, no matter what you're doing.

Hall of Fame pitcher Bob Feller, 92, is in hospice care in Cleveland. The former Cleveland Indians star is battling leukemia.

Dec 9, 2010 – Hall of Fame pitcher Bob Feller is in hospice care for the terminally ill, the Cleveland Indians announced late on Wednesday evening. According to a report on the team Web site, Feller recently was admitted to the Cleveland Clinic because of a bout with pneumonia, though he’s also fighting leukemia.

It’s the latest health setback for the 92-year-old former Cleveland Indians hurler, who was diagnosed in late August with acute myeloid leukemia. this resulted in him receiving multiple chemotherapy shots on a daily basis, according the the Cleveland Plain Dealer. At the time, he told reporters at an Indians game: “I’m just trying to be practical about it. It’s curable — but not always. only time will tell. The prognosis is: So far, so good.”

The next month, Feller had a pacemaker installed and also experienced vertigo.

Feller is the longest-tenured living member of the Baseball Hall of Fame, having been inducted in 1962. “Rapid Robert” spent his entire career with the Indians, pitching 18 seasons and going 266-162. he missed three years — 1942, ’43 and ’44 — to serve in World War II, but came back to pitch 12 more seasons.

GAO softens for-profit college report

For-profit college recruiters lied about potential earnings and deceived applicants about loans, charged undercover investigators in a Government Accountability Office report released Aug. 4.  Now the GAO has changed key passages, reports the Washington Post. all the changes make the for-profit recruiters look better.

For example, the original report charged a recruiter with telling the investigator that some graduates in architectural and civil drafting earn $120,000 to $130,000 a year. The GAO notes that 90 percent of drafters earn less than $70,000. in the revision, the recruiter adds that in the current economic climate, starting pay is only $13 to $14 an hour, $15 an hour if a graduate is “lucky.”

In another case, the original report said a recruiter had inflated salaries for massage therapists. The revision admits the recruiter gave an accurate figure, but added that the school’s massage instructors make much more.

The original report charged a recruiter had told the investigator that he’d finish the seven-month course before the details on the loan application would be checked by the Education Department. That suggestion came from the investigator, the revised report says. The recruiter merely agreed.

Undercover GAO investigators posed as prospective students in encounters with college representatives that were captured in audio and video recordings. The GAO is a nonpartisan investigative arm of Congress.

Its widely reported findings were a major political setback for the industry, and executives apologized for incidents that put their schools in an embarrassing light. Industry critics said the report buttressed their case as they pushed for a new rule requiring that for-profit colleges demonstrate that their courses lead to “gainful employment” for their students or lose access to lucrative federal student aid programs.

Democrat Sen. Tom Harkin, chair of the Health, Education, Labor and Pensions committee, used the GAO report to lambaste for-profit higher education in hearings on regulating the industry. while the GAO stands by its initial conclusion — some for-profit recruiters provide misleading information and encourage loan fraud — Republican Sen. Mike Enzi, the ranking Republican on the committee, wants the GAO to explain the changes. He wrote in a letter Tuesday to Gene L. Dodaro, the acting head of the GAO, that the “substantial” revisions raise “a number of troubling questions” and “undermine many of the allegations” in the GAO report.

Under attack in the Senate and facing tougher regulation of student loans, for-profit higher education companies have seen stock prices fall sharply. Both Apollo Group, which owns the University of Phoenix, and Kaplan have adopted new policies to screen out less-motivated students, who are unlikely to complete a program and earn enough to pay back their loans. Expecting enrollment declines, both companies are laying off employees.

Who will investigate the GAO? asks Jonathan Robe on the College Affordability blog.

Now I know that nobody is perfect and therefore the need for revision doesn’t necessarily imply malfeasance on the part of the GAO, but when you’re doing an investigation purporting to show misleading advertising and even fraud, the first rule of thumb is that you do all that you can to avoid committing the same error you accuse others of committing.

We used to live in Brechin, Angus, Scotland and we used to go to an Indian restaurant in Montrose, Scotland and order Lamb Biryani. It was a rice pilaf served with a lamb and vegetable currry that came in a separate dish and then we put it over our rice pilaf. It was very juicy and delicious and sometimes there were different vegetables in the curry, but it was all great. then we moved to the US and when we went to Indian restaurants over here, the Lamb Biryani here is just a rice and meat dish that is quite dry and has no other sauce with it. Does anyone have any idea what we were really being served in the Montrose restaurant? I have checked all sorts Indian recipes and none of them even come close. anyone have any ideas?

i dunno. but i really like Jaipur vegetables by the company "Tasty Bite"…. you ought to be able to find it in the ethnic section at a supermarket. it's a vegetable curry. It would probably taste pretty good poured over a biryani. They also make a nice Peanut Sauce with Noodles.

Maybe your restaurant started with some ghee and spices in a skillet, and then added flour to make a roux?

Lamb biryani is spicy dish with lamb in dry rice and served separately with sauce made of yogurt and onions and thin lamb gravy. This is how it is served in India.

All nations have their own interpretation of Indian dishes.
For example,a traditional Indian restaurant `in` India would probably scoff at or not know what a chicken tikka massalla is!
It,and most other dishes including your biryani has evolved to suit british tastes.
Myguess at what you are being served in the U.S.is a Pathia or Korahi but i think it is `their` interpretation of the biryani.
Sadly you will have to move back to blighty if you want a REAL RUBY!

Authentic Indian Lamb Biryani is a dry rice and meat dish (the cooking process is laborious and complex).

What you are describing is a type of Lamb Curry with vegetables served over rice, this is definitely not lamb biryani!

New iPhone app creates Barack Obama Hope style posters - iPod/iPhone - Macworld UK

The app is based on the Barack Obama iconic ‘Hope’ poster designed by artist Shepard Fairey, which was later acquired by the Smithsonian Institution for its National Portrait Gallery.

Fairey found the Obama photograph using Google Image Search, which eventually revealed to be an April 2006 photo by freelancer Mannie Garcia for the Associated Press. the image was later the centre of copyright issues, an issue that was eventually resolved.

Urban Meyer Resigns To Focus On Family

GAINESVILLE, Fla. — Florida coach Urban Meyer said he is leaving one of the premier jobs in college football for the second time to spend more time with his family.in a campus news conference Meyer said he wants to make being a husband and father his top priority.”I have not seen my two girls play high school sports. I can’t get that time back,” he said.The 46-year-old coach led Florida to two national titles but briefly resigned last December, citing health concerns, but returned the next day. He had been hospitalized with chest pains after the Gators lost to Alabama in last season’s Southeastern Conference championship game.”Last year was a knee-jerk reaction,” Meyer said. “This year was just completely different.”Florida athletic director Jeremy Foley said he had no “second guesses” about how he handled Meyer’s very brief resignation last year.”He’s at peace with his life,” Foley said. “He wasn’t at peace a year ago, and this institution helped him get there.”Meyer called Foley on Saturday to tell him he was contemplating retirement. they met Tuesday to finalize his intentions.”He’s put his heart and soul into college football,” Foley told The Associated Press before the news conference. “He’s not sick. This is a totally different situation than a year ago. He just wants to take a step back and spend time with his family.”Foley said the coaching search will begin immediately and hopes to have a new coach in the next 2&frac12; weeks.Meyer’s announcement caught players, fans and the rest of college football by surprise.Meyer called assistant coaches, many of whom were on the road recruiting, earlier this week to relay the news. He returned for spring practice in March, but managed to take significant time off before and after.He went to Hawaii with his wife, traveled to Rome and saw the pope, took a trip to Israel, visited the Masters golf tournament with his daughter and took in a Tampa Bay Rays baseball game.He said it was the first offseason in which he stepped away for days at a time.but this season he had to replace Tim Tebow and several other stars who had gone on to the NFL, and the Gators struggled mightily.Florida finished 7-5, the worst record of Meyer’s 10-year head coaching career, which began at Bowling Green. House and Senate Democratic leaders have planned an evening vote Wednesday to advance legislation to legalize young immigrants, over the opposition of most Republicans and several in their own party.

Critics call the so-called Dream Act backdoor amnesty for lawbreakers because it has no cap or end-date and allows applicants to have committed up to two misdemeanor crimes.

According to Foxnews.com, the memo lists assault, domestic violence, sexual abuse, reckless driving, and various types of fraud as among those crimes that would not disqualify applicants.

The legislation would give illegal immigrants brought to the United States before the age of 16, and who have been here for five years and graduated from high school or gained an equivalency degree, a chance to gain legal status if they joined the military or attended college.

The Congressional Budget Office has estimated that one version of the bill that applies to immigrants aged 35 and under would let more than 1 million apply for legal status over the next 10 years, and potentially allow 500,000 to receive it. A newer version of the bill only applies to those under 30, which supporters say would limit it to 300,000 or so.

Republican lawmakers also have concerns about the economic effects of the bill.

“The bill subsidizes education for illegal immigrants, grants them amnesty, encourages more illegal immigration and inevitably takes jobs from American workers. In short, the Dream Act is a nightmare for the American people,” Rep. Lamar Smith, R-Texas, said in a written statement Wednesday. “American voters want to see our immigration laws enforced, not ignored.”

Some Hispanic activists are as much against the legislation, as those who are for it. Carmen Morales, Latino activist and a leader of the Progressives for Immigration Reform, along with members of his coalition, attended a protest against the legislation in New Jersey Wednesday.

“[Latino leaders] are here to send a strong message to Congress that as Latinos, we stand staunchly opposed to any measure that puts Americans at a disadvantage and rewards illegal behavior," Morales said.

Chiefs' Cassel has appendectomy; no word on Sunday status

KANSAS CITY, Mo. — Quarterback Matt Cassel, a key reason the Kansas City Chiefs have emerged as contenders and lead the AFC West late in the season, underwent an appendectomy Wednesday.

The Chiefs said the procedure was a success and they expected Cassel to return to work this week. they did not say he would be ready to play Sunday when the Chiefs (8-4) travel to San Diego for a showdown with the Chargers (6-6), who trail Kansas City by two games in the division.

Brodie Croyle, who is 0-9 as an NFL starter and has not appeared in a game all year, took most of the practice snaps Wednesday.

The Chiefs also signed quarterback Tyler Palko to the 53-man roster from the practice squad.

Coach Todd Haley, who insisted on saying during his news conference only that Cassel had “an illness,” declined to speculate on whether Cassel will be ready for Sunday’s game.

“Again, he’ll be on the injury report as illness today,” Haley said. “And we’ll just take it from there as we do all injuries.”

In his second season as the Chiefs’ full time starter, Cassel has been thriving under the tutelage of Haley and first-year offensive coordinator Charlie Weis, connecting on 212 of 354 passes for 2,503 yards and 23 touchdowns. He’s thrown an NFL-low four interceptions. his current streak of 158 passes without an interception is the third-longest active streak in the league.

Cassel was the AFC’s Offensive Player of the Month in November, completing 90 of 144 passes for 1,111 yards and 12 TDs with only one interception.

Steelers quarterback Ben Roethlisberger underwent an appendectomy four days before the 2006 NFL opener. he returned to play in Pittsburgh’s Week 2 game 15 days after the surgery, and his passer ratings in that and the following game — 38.7 and 30.7 — are among the six worst of his career.

Croyle, a third-round pick out of Alabama in 2006, was projected as the Chiefs’ foundation quarterback before Herman Edwards was fired as head coach after the 2008 season. Injury prone, he to the second team when new general manager Scott Pioli traded for Cassel before the 2009 season. Croyle has not started since the 2009 season opener, when Cassel was hurt.

Croyle said this week will be no different than any other for him, other than the fact he’ll be getting more practice reps.

“Just got to prepare like you always do,” he said. “My number’s called, got to be ready to go. You’ve got to be ready to go on any play.”

Croyle has seen action in 16 regular-season games, completing 173 of 300 passes for 1,631 yards, eight touchdowns and eight interceptions.

The Chiefs also placed cornerback Jackie Bates on injured reserve and signed linebacker Micah Johnson to the practice squad.

Issei Sagawa: The Man Who Made a Living Off A Brutal Murder

Issei Sagawa is a minor celebrity in Japan, but he didn’t find fame through a legitimate practice. rather, he became well known for a brutal crime he committed while a student in Paris, France.

In 1981, Sagawa murdered and cannibalized a Dutch classmate Renee Hartevelt while a student at the the prestigious Sorbonne. in interviews, he had said he chose her because of her "health and beauty," characteristics he felt he lacked. He believed he could "absorb her energy" by cannibalizing her.

Sagawa attempted to dump her body in a remote lake, but was caught in the act and then arrested by French police. having been born into a wealthy family, Sagawa’s father found a top lawyer for his defense.

For two years, Sagawa spent time in jail, and without trial, a judge found him legally insane. He was then sent to a mental institute, where he was interviewed by an author and became the subject of the book In the Fog.

Because of the book, publicity was generated about the killer and he became a macabre celebrity. French authorities decided to extradite him to Japan, where psychologists found him to be sane, but "evil" and aware of his crimes. However, because proper paperwork from French authorities did not arrive, they could not legally hold him, and he was let go.

Ok so let me go through my Orexigen vote estimate. I'll try to give a brief description next to each panel member and why I think they will vote the way they will. at the end of the analysis I give my final panel estimate. Abraham Thomas- I think Dr. Thomas will vote no. He had noted at Qnexa's panel hearing that, "The current medications available are not very effective and have a lot of side effects. the sponsors did an outstanding job of proving the efficacy, and this medication in terms of efficacy is far superior to anything that’s on the market. the concerns we have are with safety." He then went on to list a number of concerns, one of them being patients that have a history of Myocardial Infarction and the fact that on Qnexa MI's happened only on their drug. Which just happens to happen to 3 patients on Contrave and none on placebo as well, so I don't see this doctor doing a change up for Contrave, especially given the much weaker efficacy of the drug.

As anyone who’s tuned in to Celebrity Rehab knows, the post-rock lifestyle isn’t always a smooth run for once-famous musicians. now, the man partly responsible for a slew of wedding staples your grandma loves to get down on has hit a bad patch and is heading for rehabilitation, but not the kind you can milk for a last 5 minutes of fame on a reality TV tearjerker.

Richard Finch, a founding member and former bass player with KC and the Sunshine Band, has been found guilty of charges related to the sexual abuse of boys in Licking County, according to the Columbus Dispatch.

The 56-year old was looking at counts of unlawful sexual conduct with a minor, importuning and sexual imposition. He pleaded no contest. a judge found him guilty.

Finch was first nabbed by police in March. at the time he was running a home recording studio where he worked with “young, aspiring musicians.”

His victims — seven boys — were reportedly regulars at the studio. Officials say Finch told his victims they “would need to be prepared for such activity if they sought success in the music industry.”

In court Finch was allegedly contrite, saying the arrest was “a wake-up call.” (Duh). He was given a seven year prison sentence, a legal slap lightened by the prosecution’s belief a dependence on alcohol was responsible for Finch’s behavior.

In addition to sexual abuse, Finch is also responsible for some truly shitty music. Alongside partner-in-crime Harry “KC” Casey, he founded the Sunshine Band in 70s Miami (cocaine) and subsequently churned out vanilla funk hits like “get down Tonight” and “(Shake, Shake, Shake) Shake your Booty,” — titles that — in light of recent developments — are just disturbing. According to Finch’s Wikkipedia page, KC and he had a business dispute in the early 80s, precipitating the bass player’s exit from the band. KC still tours with a new line-up, and when Finch was arrested threw up a post on his web site distancing himself from his former bandmate.

I have a Kyocera Strobe, and the A button and the Left Arrow on the QWERTY keyboard don't always work. I was wondering if they'll repair it or fix it since they don't make my phone model anymore. I was wondering what is covered under a U.S. Cellular warranty… if I would be able to 'help' my phone break, and how I would want to 'help' it along a path of broken-ness… if anyone knows, do tell!

Scientists Ratchet Up Understanding of Cellular Protein Factory

ScienceDaily (Dec. 3, 2010) — Theoretical biologists at Los Alamos National Laboratory have used a new Mexico supercomputer to aid an international research team in untangling another mystery related to ribosomes — those enigmatic jumbles of molecules that are the protein factories of living cells. the research, published December 2 in the journal Nature, could aid in development of new antibiotics used to fight multidrug resistant superbugs such as MRSA (methicillin-resistant Staphylococcus aureus infections) found in many U.S. hospitals. the work may also be important for combating engineered strains of anthrax and plague.

In the context of synthetic biology, understanding the ribosome could be key to developing nanofactories that produce designer biomolecules and polymers.

In the paper, "Head swivel on the ribosome facilitates translocation via intra-subunit tRNA hybrid sites," Los Alamos National Laboratory researchers Karissa Sanbonmatsu and Paul Whitford and José N. Onuchic at the University of California-San Diego join Christian Spahn, Andreas Ratje, and others from the Institute for Medical Physics and Biophysics, Berlin, Germany, to describe for the first time how a complicated swivel movement within a bacterial ribosome accommodates synthesis of proteins.

Ribosomes are composed of long chemical chains, called ribonucleic acids (RNA), and proteins. each ribosome has two interlocked subunits, one large and one small, which behave as a single molecular machine. Because of its makeup, each ribosome resembles a tangle of threads or a handful of rubber bands tossed together. despite the ribosome’s outwardly disjointed appearance, researchers have found that the two subunits ratchet, un-ratchet, and swivel during protein synthesis to allow introduction of helper chemicals called transfer RNAs (tRNAs) into its folds to manufacture new chains of protein molecules. the proteins are used to create new cells or perform necessary functions within the host cell or organism.

Ribosomes build proteins by linking chemical segments fashioned from instructions delivered via messenger RNA, which is DNA’s molecular cousin. each segment, or amino acid, corresponds to a trio of bases in the message that, in turn, complement trios encoded in transfer RNA. each base in the trio corresponds to a single chemical complement found on the RNA. In order for protein synthesis to occur, the tRNA must bind to the ribosome at two distinct sites — one to decode the information and another to link the new amino acid to the emerging protein.

After each amino acid is added, the ribosome must crawl along the message to create additions. Exactly how this crawling occurs has been a mystery for several decades. Researchers have suspected that ratcheting motions of the two ribosomal subunits are key to allowing RNA and associated catalysts into the complex structure of the ribosome so the RNA and ribosome can couple at the crucial sites to create proteins. In the Nature paper, the researchers discovered that the majority of crawling (movement along messenger RNA) occurs during a new kind motion, "head swivel," rather than ratcheting.

The paper describes how an antibiotic was used to inhibit the full swivel and ratcheting motion of a ribosome from a bacterium called Thermus Thermophilus, which thrives in hot acidic environments. the ribosomes were flash-frozen at various mid-swivel and mid-ratchet configurations and examined under a powerful electron microscope.

The observed configurations were then coupled with a computer model newly developed at Los Alamos called MDFIT. the computer algorithm integrates molecular simulation with maps of ribosome structures obtained through the cryogenic microscopy. the Los Alamos team then used the Encanto supercomputer — funded by the state of new Mexico and housed at the Intel plant in Rio Rancho — to create molecular snapshots of the complicated motion of the ribosomal subunits during protein synthesis.

Previously, scientists were only able to observe the beginning or end states of the motion. These new images show the behavior of the ribosome through its range of motion — much like early photographic motion studies that showed the entire fluid movement of a galloping horse. In addition to showing the importance of head swivel motion, the study showed that a key catalyst in the process acts as a dynamic pawl in the ribosomal machinery, providing directionality and acceleration for translocation of the tRNA. the understanding provided by the new model will help researchers to develop more effective antibiotics that target the ribosomal machinery of harmful organisms.

"while static images of the ribosome have revealed the detailed structure of the complex, we still don’t know how all the parts of the machine work together to make proteins," said Janna Wehrle, Ph.D., who oversees Dr. Sanbonmatsu’s and other structural biology grants at the National Institutes of Health. "by showing how the bacterial ribosome carries out a key step of protein synthesis, this study has begun to produce a more dynamic picture while offering a new way to target harmful, multi-drug resistant bacteria."
